Amount of Lectin Required for Primary Global Screening Protocol: Participating Investigator (PI) supplies Core H with 1 to 2 mg purified lectin
Number of unique ligands: 115
Replicates: 3
Positive Controls: 3
Negative Controls: 3
Volume per well: 50ml
Total volume lectin required: 17.25ml
Lectin concentration: 30mg/ml
Total FITC-labeled Lectin or Igchimera required per primary screening: 518mg
 
FITC-Labeling of Lectins: Alexa Fluor 488 Protein Labeling Kit (A-10235) from Molecular Probes
Minimum requirement: 1mg/0.5ml
Lectin labeled and purified per manufacturer’s protocol
 
Ig-chimera Analysis Using FITC-Labeled Secondary Antibodies:
 
PI specifies primary host or Ig used
Core H will provide Alexa-labeled secondary antibody
 
Special Conditions: Participating Investigators are to contact Rick Alvarez, Director of Core H , prior to sending lectins for analysis [special considerations on a case by case basis]
