Cortical organoids have emerged as a transformative platform in modern neuroscience, offering an
unparalleled model of human cortical development that 2D cultures and animal models cannot provide. Yet,
the tools to rigorously interrogate organoids network dynamics lag behind the biology. We present custom
transparent and flexible microelectrode arrays that enable simultaneous electrophysiological recording
and imaging, purpose-built for organoid research both in vitro and in vivo. Using this platform, we
reveal how spontaneous and stimulated networks evolve in organoids, and how implanted neural tissue
functionally integrates with host circuitry, opening new frontiers in neural prosthetics, personalized
medicine, and biological computing.
