Best Sorting Practices

  • Concentrate cells to optimal #’s of 50-100 million cells/ml (Lower concentrations can be run at slower rates)
  • Lymphocytes-concentrate 50 million cells/ml
    Sticky/Adherent cells-concentrate 20 million cells/ml

  • Filter your samples using Falcon 12x75mm polystyrene test tubes (Falcon #2235)
  • Minimum sample volumes= 0.5 ml

Maximum sample volume= 3.5 ml

  • Bring controls
    • Unstained
    • Single color controls
  • Bring collection tubes precoated w/100% FBS or 5% BSA. Use 1ml of media + 20% serum or 1ml of 100% serum
  • Bring extra collection tubes
  • If collecting into plates, add 100 ul of media into wells

How to get cells not to be clumpy for sorting or analysis:

1 mM EDTA sticky cells
10 units DNAse/ml lots of dead cells
